Knowing that sign sits behind signal, signature, significant and designate lets a reader make a reasonable first guess at any of them. Derivations change a word's part of speech as well as its meaning: adding -ion turns an action into a thing, and adding -ly turns a description into a manner. Meaning still has to be confirmed in context, because words drift away from their roots over time.
